Planning to travel to Mexico in 2020 for a friends destination wedding. Full cost of trip must be paid 4 months in advance. I am only able to request vacation from work AFTER the trip will have been paid, and there is no guarantee I will be granted the time off. Certain providers offer a Missed Connection benefit for these situations, although most of the providers only offer this if you miss a cruise or tour departure. MH Ross and AXA Assistance USA do not specify that it has to be a tour or cruise departure that is missed due to an airline delay. The hotels I have booked are allowed free cancellation 3 - 7 days before my stay with no fees charged. If I need to interrupt my trip and end up being charged by the hotel because free cancellation period has passed, will trip interruption cover the expenses?

You can only insure the cost of the hotel if its is prepaid and non-refundable prior to departure. If you are not prepaying for the hotel prior to departure, you are not going to be covered for Trip Interruption, unfortunately.

I have a nonrefundable roundtrip air ticket. If I need to change the return flight, the airline will give me a credit toward another flight, minus a $200 change fee. Therefore, although I must insure the entire cost of my roundtrip ticket, my actual loss will only be $200 (plus additional cost of the new ticket, if any). Will trip interruption coverage at 150% reimburse me $300 for the change fee?

It would depend on the plan, since they would all be worded differently. But usually, when there is 150% Trip Interruption, they reimburse for the unused portion of your prepaid, non-refundable trip costs and reimburse for the additional transportation expenses incurred to reach the return destination due to one of the covered reasons. You can click on the amount next to Trip Interruption when viewing a plan to read the details of that specific plan.
